Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.

Development of the compound took place in the Soviet Union during the 1980s, largely through peptide research groups affiliated with the Shemyakin-Ovchinnikov Institute and Moscow State University. Investigators screened fragments of adrenocorticotropic hormone for activity on the central nervous system while attempting to separate cognitive effects from hormonal ones. The shortened sequence was selected because it retained behavioral activity in animal models without stimulating corticosteroid release. Early publications described the molecule as an ACTH(4-10) analog.
Published research on this peptide originates mainly from Russian laboratories, and the wider international literature is comparatively thin. Studies have used rodent models of stroke, hypoxia, and memory tasks, with a smaller number of human trials reported. Reported outcomes include changes in attention and memory measures, along with effects on neurotrophic factor expression in some experiments. Small sample sizes, inconsistent dosing protocols, and limited independent replication are widely noted limitations, so the strength of the clinical evidence remains an open question.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. The first four residues correspond to the ACTH(4-10) fragment, while the terminal Pro-Gly-Pro extension was added to improve metabolic stability. Its molecular formula is C37H51N9O10S and its monoisotopic mass is approximately 813.9 Da. The compound is classified as a peptide research chemical and as a registered pharmaceutical product in Russia, but it does not hold marketing approval in the European Union or the United States. Naming conventions vary across supplier catalogs.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.

== Biography == Leonard Thompson was born on Pickering Street near the Beaches neighbourhood of Toronto on 17 July 1908, to parents Harold and Florence Thompson. He was diagnosed with diabetes mellitus, and was first treated at the Hospital for Sick Children before being transferred to the care of physicians Andrew Almon Fletcher, Duncan Archibald Graham, and Walter Ruggles Campbell. Thompson received his first injection in Toronto, Ontario, on 11 January 1922, at 13 years of age. Thompson's first dose had an apparent impurity which caused an allergic reaction. A refined process was quickly developed to concentrate the pancreatic extract. Twelve days later, on 23 January, he began a two-week series of daily injections that rapidly improved his health, allowing him to live until dying of bronchopneumonia at age 26. Until insulin was made clinically available, a diagnosis of diabetes was a death sentence, more or less quickly (usually within months, and frequently within weeks or days).
== Function == Actin's primary role in the cell is to form linear polymers called microfilaments that serve various functions in the cell's structure, trafficking networks, migration, and replication. The multifaceted role of actin relies on a few of the microfilaments' properties: First, the formation of actin filaments is reversible, and their function often involves undergoing rapid polymerization and depolymerization. Second, microfilaments are polarized – i.e. the two ends of a filament are distinct from one another. Third, actin filaments can bind to many other proteins, which together help modify and organize microfilaments for their diverse functions. In most cells actin filaments form larger-scale networks which are essential for many key functions:
Antihypertensive drugs Anti-anxiety drugs (e.g. benzodiazepines) Antipsychotic drugs Anticonvulsants Non-steroidal anti-inflammatory drugs (e.g. Antihistamine) Opioid Medical marijuana Recreational drugs
The two substrates of this enzyme are scytalone and oxidised nicotinamide adenine dinucleotide phosphate (NADP+). Its products are naphthalene-1,3,6,8-tetrol, reduced NADPH, and a proton. This enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-OH group of donor with NAD+ or NADP+ as acceptor. The systematic name of this enzyme class is scytalone:NADP+ Delta5-oxidoreductase.

After the defeat of Germany in World War I, the Allied powers arranged a plebiscite in northern and central Schleswig. The plebiscite was conducted under the auspices of an international commission, which designated two voting zones to cover the northern and south-central parts of Schleswig. Steps were taken to also create a third zone covering a southern area, but zone III was cancelled again and never voted, as the Danish government asked the commission not to expand the plebiscite to this area. In zone I covering Northern Schleswig (10 February 1920), 75% voted for reunification with Denmark and 25% voted for Germany. In zone II covering central Schleswig (14 March 1920), the results were reversed; 80% voted for Germany and just 20% for Denmark. Only minor areas on the island of Föhr showed a Danish majority, and the rest of the Danish vote was primarily in the town of Flensburg. On 15 June 1920, Northern Schleswig officially returned to Danish rule. The Danish/German border was the only one of the borders imposed on Germany by the Treaty of Versailles after World War I that was never challenged by the Nazis. In 1937, the Nazis passed the so-called Greater Hamburg Act (Groß-Hamburg-Gesetz), where the nearby Free and Hanseatic City of Hamburg was expanded, to encompass towns that had formerly belonged to the Prussian province of Schleswig-Holstein.
=== Smoking cessation === Cytisine has been available in post-Soviet states as an aid to smoking cessation under the brand name Tabex from the Bulgarian pharmaceutical company Sopharma AD. In 1961, Bulgarian pharmacist Strashimir Ingilizov synthesized Tabex using the alkaloid Cytisine which was derived from the seeds of the yellow acacia (Cytisus laburnum), a European decorative shrub prevalent in Bulgaria and commonly referred to as "golden rain". It was first marketed in Bulgaria in 1964 and then became widely available in the Soviet Union. In Poland, it is sold under the brand name Desmoxan, and it is also available in Canada under the brand name Cravv. In June 2026, the U.S. Food and Drug Administration issued a complete response letter for Achieve Life Sciences' application to market cytisinicline for smoking cessation, citing deficiencies at a third-party manufacturing facility and unfinished product labeling. Its molecular structure has some similarity to that of nicotine, and it has similar pharmacological effects. Like the smoking cessation aid varenicline, cytisine is a partial agonist of nicotinic acetylcholine receptors (nAChRs). Cytisine has a short half-life of 4.8 hours. As a result, the extract provides smokers with satisfaction similar to smoking a cigarette, alleviating the urge to smoke and reducing the severity of nicotine withdrawal symptoms, while also reducing the reward experience of any cigarettes smoked.
==== Metabolic and endocrine ==== Abnormal thyroid function may result in the development of catatonia when the thyroid overproduces (hyperthyroidism) or underproduces thyroid hormones (hypothyroidism). This is thought to occur due to the impact of thyroid hormones on metabolism, including in the cells of the nervous system. Abnormal electrolyte levels have also been shown to cause catatonia in rare cases. Most notably, low blood sodium levels can cause catatonia in some people.
===== China ===== Indian concerns over the threat of China led to collaboration with the United States in the first two Cold War decades. However, American support for Pakistan in the Indo-Pakistani war of 1971 and the 1972 visit by Richard Nixon to China marked a turning point in the triangular relationship toward stronger China–United States relations, so India pivoted to the Soviet Union for military support. In the 21st century, India-United States cooperation has aimed to contain China in Asia and the broader Indo-Pacific.

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
